Single-cell RNA sequencing has become the logical choice to overcome the cellular heterogeneity in tissue samples. However, spatial organization and localization of the transcript is necessary for a better understanding of pathophysiological conditions.
In situ sequencing allows us to sequence RNA transcripts directly in a fixed section or cells with sub-cellular level spatial information.
The method uses nucleic acid bases labeled with unique fluorescent dyes, padlock probes for target RNA’s, and the enzymes that catalyze the formation of circularized DNA for rolling circle amplification.
